I come from two perspectives here: as a rank amateur machinist (my lathe has yet to be plugged in!) and as a moderately knowledgeable benchrest shooter. So here goes... Gordy and the folks from Grizzly have put together an exceptional video outlining a sequence of steps that will produce a championship chamber job. For through-the-headstock chambering, this is the method. Now, this video is partly a promo for the Grizzly lathe so the exact technique demonstrated is valid for machines with a narrow headstock (not mine), but the critical steps shown are easily re-interpretted for whatever style of 'smithin' you may have to do based on your equipment.
